Overview

DS-8895a is a humanized, afucosylated IgG1 monoclonal antibody that targets the ephrin type-A receptor 2 (EphA2), a receptor tyrosine kinase overexpressed in various solid tumors. By binding to EphA2 on tumor cells, DS-8895a induces potent antibody-dependent cellular cytotoxicity (ADCC), leading to targeted tumor cell death. The afucosylation of the antibody enhances its ADCC activity by increasing affinity for FcγRIIIa/CD16 on natural killer cells. Developed primarily for the treatment of advanced solid tumors such as gastric and esophageal cancers, clinical trials demonstrated manageable safety but limited efficacy, with some patients achieving stable disease or partial response. The drug was developed by Daiichi Sankyo and reached phase I clinical trials before discontinuation[1][2][3][4][5][8].
