Overview

DS1040b is a novel small molecule inhibitor of the activated form of thrombin-activatable fibrinolysis inhibitor (TAFIa). By inhibiting TAFIa, DS1040b enhances endogenous fibrinolysis, potentially improving clot breakdown without significantly increasing bleeding risk. It was developed as a potential therapy for thromboembolic diseases such as acute ischemic stroke and pulmonary embolism. Clinical studies have shown that DS1040b substantially decreases TAFIa activity and shortens clot lysis time in a dose-dependent manner, with no significant impact on bleeding time or coagulation parameters observed in early-phase trials. The drug has been evaluated primarily via intravenous infusion in both healthy volunteers and patients with thromboembolic conditions[5][6][9].
