Overview

DS16570511 is a potent and selective small molecule inhibitor of the mitochondrial calcium uniporter (MCU), a channel responsible for calcium uptake into the mitochondrial matrix. Developed by Daiichi Sankyo, the compound is cell-permeable and functions by blocking mitochondrial calcium overload, which is a key driver of mitochondrial dysfunction and reactive oxygen species (ROS) production. In preclinical studies, DS16570511 has been shown to attenuate heme-induced platelet activation and the release of pro-thrombotic factors like thrombospondin-1 (TSP-1), suggesting potential therapeutic utility in hemolytic disorders such as sickle cell disease. It is also widely utilized as a pharmacological tool to investigate the role of mitochondrial calcium signaling in cardiovascular diseases and ischemia-reperfusion injury.
