DSP-0187 is an investigational small molecule orexin 2 receptor (OX2R) agonist developed by Sumitomo Pharma for the treatment of narcolepsy. Orexins (also known as hypocretins) are essential neuropeptides that regulate wakefulness and the sleep-wake cycle; a deficiency in orexin signaling is the primary underlying cause of Narcolepsy Type 1. By selectively targeting and activating the OX2R, DSP-0187 aims to mimic the action of endogenous orexin, thereby promoting wakefulness and potentially reducing symptoms such as excessive daytime sleepiness (EDS) and cataplexy. The compound has been evaluated in early-stage clinical trials, including Phase 1 ascending single oral dose studies in healthy volunteers, to assess its safety, tolerability, and pharmacokinetic profile.
