Overview

DSP-0337 is an orally bioavailable small molecule inhibitor of ubiquitin-specific protease 7 (USP7), currently under clinical investigation for the treatment of advanced solid tumors. USP7 is a deubiquitinating enzyme that plays a critical role in regulating the stability of several key proteins involved in tumor suppression and oncogenesis, most notably the MDM2-p53 axis. By inhibiting USP7, DSP-0337 promotes the degradation of MDM2, which in turn leads to the stabilization and activation of the p53 tumor suppressor protein, inducing cell cycle arrest and apoptosis in p53-wildtype cancer cells. Additionally, USP7 inhibition may modulate the immune microenvironment by affecting regulatory T cell (Treg) function. The drug is being developed by Sumitomo Pharma America (formerly Sumitomo Dainippon Pharma Oncology).
