DSP-0509 (guretolimod) is a small molecule agonist of Toll-like receptor 7 (TLR7), designed to stimulate innate immune responses within the tumor microenvironment. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, functioning as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor activity. The combination of DSP-0509 and pembrolizumab aims to synergistically activate both innate and adaptive immunity, potentially converting immunologically "cold" tumors into "hot" tumors that are more responsive to immunotherapy. This combination has been investigated in phase 1/2 clinical trials for patients with advanced solid tumors, with Sumitomo Dainippon Pharma Oncology as the developer[1][4][6][8].
