Overview

DSP-1053 is a novel benzylpiperidine derivative developed as an antidepressant. It acts as a potent serotonin transporter (SERT) inhibitor and also exhibits partial agonist activity at the 5-HT1A receptor. This dual mechanism enhances serotonergic neurotransmission, which is associated with antidepressant effects. Preclinical studies demonstrated that DSP-1053 increases extracellular serotonin levels and produces rapid antidepressant-like effects in animal models of depression, with fewer undesirable side effects such as nausea and emesis compared to conventional selective serotonin reuptake inhibitors (SSRIs). The drug was developed by Sumitomo Pharma for the oral treatment of depressive disorders but its development has been discontinued[1][2][3][4][8].
