Overview

DSP-3905 is a selective antagonist of the voltage-gated sodium channel Nav1.7 (SCN9A), originally developed by Sumitomo Pharma and subsequently in-licensed by AlphaNavi Pharma in October 2023. Nav1.7 is highly expressed in peripheral sensory neurons and plays a critical role in the transmission of pain signals; gain-of-function mutations in the SCN9A gene are associated with severe pain disorders, while loss-of-function mutations lead to congenital insensitivity to pain. DSP-3905 is designed to provide potent analgesia for neuropathic pain conditions while minimizing off-target effects on other sodium channel subtypes, such as Nav1.5 in the heart or Nav1.1/1.2 in the central nervous system. The compound has completed early clinical evaluation, including Phase 1 single- and multiple-ascending dose studies conducted in the United States.
