Overview

DT-7012 is a differentiated anti-CCR8 monoclonal antibody developed for cancer immunotherapy. It selectively depletes tumor-infiltrating regulatory T cells (Tregs) within the tumor microenvironment by engaging antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). By targeting CCR8, a chemokine G-protein coupled receptor highly expressed on immunosuppressive Tregs, DT-7012 transforms the tumor microenvironment into an immunocompetent state, enhancing anti-tumor immune responses. The drug is designed to bind CCR8 with high affinity regardless of post-translational modifications or ligand presence and prevents CCL1-induced internalization of CCR8, ensuring sustained depletion of Tregs even in challenging conditions. This mechanism aims to overcome resistance seen with other immunotherapies and benefit patients who do not respond to existing treatments. DT-7012 is being developed by Domain Therapeutics and has demonstrated strong preclinical efficacy as a monotherapy in cancer models[1][5][6][7].
