DT-9046 is a highly differentiated, first-in-class, orally bioavailable small molecule that acts as a biased negative allosteric modulator (NAM) of Proteinase-Activated Receptor 2 (PAR2). Developed by Domain Therapeutics, DT-9046 represents a novel approach to treating multiple inflammatory diseases—including atopic dermatitis, inflammatory bowel disease, arthritis, and neuroinflammatory conditions such as migraine. Its unique mechanism targets an allosteric binding site on PAR2 to fine-tune GPCR function and selectively modulate only the pathways critical for therapeutic effects. This strategy aims to maximize efficacy while minimizing resistance and side effects. Unlike previous PAR2-targeting strategies—mainly biologics or full antagonists—DT-9046 is the first oral drug candidate in this class to reach preclinical development[2][3][5][6].
