It acts as a positive allosteric modulator (PAM) of these group III mGluRs. The rationale for this approach is based on the presynaptic localization of these receptors in the basal ganglia, where they regulate neurotransmitter release implicated in motor symptoms and levodopa-induced dyskinesia (LID) in Parkinson’s disease. By potentiating all three subtypes simultaneously, DT095435 aims to provide pro-motor effects and reduce dyskinesia associated with long-term levodopa therapy. Preclinical studies have shown robust anti-dyskinetic activity and improvement in motor function in non-human primate models of LID[1][2][9].
