Overview

DT204 is a small molecule inhibitor of S-phase kinase-associated protein 2 (SKP2), a component of the SCF (Skp, Cullin, F-box containing) E3 ubiquitin ligase complex. SKP2 is known to promote cell cycle entry and glycolysis, and its inhibition is being explored as a strategy to preserve the integrity and regenerative capacity of hematopoietic stem cells (HSCs). In preclinical studies, DT204 has demonstrated the ability to enhance HSC quiescence, expand stem cell pools, and protect hematopoietic stem and progenitor cells (HSPCs) from genotoxic stress and irradiation-induced DNA damage during ex vivo manipulation. By modulating metabolic activity—specifically reducing oxidative phosphorylation and glycolysis—DT204 helps maintain the stemness of HSCs, making it a potential candidate for improving outcomes in bone marrow transplantation and gene therapy protocols.
