Overview

DT2219ARL is a novel bispecific ligand-directed toxin (BLT) consisting of two single-chain variable fragments (scFv) recognizing human CD19 and CD22 linked to the first 389 amino acids of diphtheria toxin (DT390). It targets cancer cells expressing the CD19 receptor, CD22 receptor, or both receptors. When DT2219ARL binds to cancer cells, they internalize the drug and are killed due to the action of the drug's cytotoxic payload. The drug has demonstrated potency in the IC50s 0.06–0.2 nM range and has shown success in early human clinical trials for patients with relapsed/refractory B-cell lymphoma or leukemia.
