Overview

DT433 is a DNAzyme therapeutic investigated for its role in targeted gene silencing, specifically focusing on the inhibition of DNMT1 expression and reactivation of tumor suppressor pathways (such as upregulation of p16 mRNA) in cancer cells. DT433 was designed as a catalytic DNA molecule (DNAzyme) to selectively cleave and silence disease-causing RNA. In preclinical studies, DT433 treatment resulted in inhibition of DNMT1, which is linked to epigenetic regulation and tumor progression, and increased p16 mRNA, a key tumor suppressor, suggesting restoration of cell cycle control in cancer models[1]. DNAzymes represent an emerging class of synthetic nucleic acid therapeutics with highly specific RNA targeting capabilities.
