DTA-1 is an agonistic monoclonal antibody that targets mouse GITR (glucocorticoid-induced tumor necrosis factor receptor, also known as TNFRSF18), a co-stimulatory immune checkpoint molecule in the tumor necrosis factor receptor superfamily. The original DTA-1 antibody is a rat IgG2b/λ, although chimeric (mouse IgG2a/κ) and murinized (mouse IgG2c/λ) versions have also been engineered to reduce immunogenicity in experimental models. DTA-1 binding to GITR promotes T cell activation and proliferation and is used extensively in preclinical immuno-oncology research to study immune modulation and antitumor responses. GITR is constitutively expressed at high levels on regulatory T cells and upregulated on activated effector T cells. DTA-1 has shown that effective engagement of Fcγ receptors is critical for its antitumor effect. The antibody is most prominent as a surrogate in vivo tool in mice and is not a human therapeutic or approved pharmaceutical product[1][3].
