DTC1 (also known as DTC-1) is a small molecule inhibitor of the Wnt/beta-catenin signaling pathway. It specifically targets the interaction between beta-catenin and the CREB-binding protein (CBP), a key transcriptional co-activator. By disrupting this interaction, DTC1 inhibits the transcription of Wnt-target genes that are involved in cell proliferation, survival, and epithelial-mesenchymal transition (EMT), which are often dysregulated in various cancers. Developed by DTC Biotech (Dalian Tongze Cancer Biotech), DTC1 is primarily being investigated for its potential in treating solid tumors, particularly colorectal cancer, where Wnt pathway mutations are highly prevalent. The drug aims to overcome resistance to conventional therapies by targeting the underlying oncogenic signaling driven by the beta-catenin/CBP complex.
