Overview
DTX-022 is an investigational antibody-drug conjugate (ADC) developed by Dantari, utilizing its proprietary DARE (Dantari Antibody-drug Conjugate platform) technology. It is designed to target HER2-positive cancers, with a specific emphasis on treating brain metastases, a significant unmet need in HER2+ breast cancer. The drug consists of a HER2-targeting antibody conjugated to a potent topoisomerase I inhibitor payload. The DARE platform is engineered to provide a high drug-to-antibody ratio (DAR) while maintaining favorable pharmacokinetic properties and enhancing the delivery of the payload across the blood-brain barrier (BBB). By optimizing the delivery of the cytotoxic agent to the central nervous system, DTX-022 aims to overcome the limitations of current HER2-targeted therapies in treating intracranial disease.
