Dual chimeric antigen receptor cytokine-induced killer cells IL-3z WT are an investigational, genetically engineered cytokine-induced killer cell product evaluated preclinically for **acute myeloid leukemia**. The cells co-express a wild-type interleukin-3 zetakine CAR that uses membrane-tethered IL-3 to recognize the interleukin-3 receptor alpha chain CD123 and deliver CD3 zeta activation signaling, together with a CD33-directed chimeric costimulatory receptor. This balanced dual-targeting strategy was studied to preferentially activate effector cells against CD123-positive and CD33-positive AML cells while addressing on-target toxicity risks associated with either antigen alone. The wild-type IL-3 zetakine construct was evaluated alongside lower-affinity IL-3 variants; published work emphasized lower-affinity constructs as the more selective approach.
