Overview

Dual-switch GoCAR-T is a cell therapy platform developed by Bellicum Pharmaceuticals designed to treat solid tumors with enhanced control over efficacy and safety. The platform incorporates two independently regulated molecular switches: a "GO" switch (iMC) and a "STOP" switch (iRC9/RapaCIDe). The "GO" switch is activated by the small molecule rimiducid, which triggers MyD88 and CD40 signaling to provide costimulation, promoting T-cell expansion and persistence in the immunosuppressive tumor microenvironment. The "STOP" switch is a rapamycin-regulated safety mechanism that induces apoptosis via caspase-9 dimerization to mitigate potential toxicities. The lead candidate using this platform, BPX-603, targets HER2-positive solid tumors, including breast, gastric, and esophageal cancers. Clinical development of the platform was terminated following Bellicum's restructuring and liquidation.
