This is an investigational, allogeneic, off-the-shelf chimeric antigen receptor natural killer (CAR-NK) cell therapy developed by Beijing Biotech for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC). The therapy utilizes NK cells derived from healthy donor cord blood, which are genetically engineered to express dual-target CAR constructs using a tandem OR-gate approach. These constructs target combinations of tumor-associated antigens, specifically mesothelin (MSLN), epidermal growth factor receptor (EGFR), and human epidermal growth factor receptor 2 (HER2). The engineered NK cells also express interleukin-15 (IL-15) to enhance in vivo survival and persistence and incorporate an inducible safety switch (iCasp9) to allow for controlled depletion of the cells if necessary. The therapy is currently being evaluated in a biomarker-guided Phase 1/2 clinical trial where patients receive a CAR-NK product matched to their specific tumor antigen profile.
