This is a combination cancer therapy that pairs **duligotuzumab** (a dual-action, humanized IgG1 monoclonal antibody targeting both epidermal growth factor receptor [EGFR] and human epidermal growth factor receptor 3 [HER3]) with two chemotherapeutic agents, **cisplatin** and **5-fluorouracil**. * Duligotuzumab blocks ligand binding and downstream signaling of both EGFR and HER3, inhibiting tumor cell proliferation and mediates antibody-dependent cell-mediated cytotoxicity (ADCC)[1][3][5]. * Cisplatin is a platinum-based chemotherapy agent that causes DNA crosslinking and cell death, especially in rapidly dividing cells. * 5-fluorouracil (5FU) is an antimetabolite that inhibits thymidylate synthase, blocking DNA synthesis in cancer cells. This regimen has been investigated in solid tumors such as recurrent/metastatic squamous cell carcinoma of the head and neck (HNSCC), and was shown to provide enhanced antitumoral effects compared to monotherapy in preclinical models and early clinical trials[1][3][7]. Duligotuzumab was developed by Genentech/Roche.
