Duloxetine + gabapentin is a combination therapy being investigated by Beni-Suef University for the treatment of chemotherapy-induced peripheral neuropathy (CIPN). Duloxetine is a serotonin-norepinephrine reuptake inhibitor (SNRI) that increases the synaptic concentration of serotonin and norepinephrine, thereby enhancing descending inhibitory pain control in the central nervous system. Gabapentin is a gabapentinoid that binds with high affinity to the alpha-2-delta-1 subunit of voltage-gated calcium channels, leading to a reduction in the calcium-dependent release of pro-nociceptive neurotransmitters. This combination approach aims to target multiple mechanisms of neuropathic pain to achieve superior efficacy compared to monotherapy in cancer patients suffering from CIPN.
