Duloxetine + olanzapine is a combination of two small molecule pharmaceuticals used together, primarily in psychiatric and mood disorder contexts. Duloxetine is a serotonin-norepinephrine reuptake inhibitor (SNRI) antidepressant that increases levels of serotonin and norepinephrine in the central nervous system by inhibiting their reuptake. Olanzapine is an atypical antipsychotic that acts as an antagonist at multiple neurotransmitter receptors, including dopamine D2 and serotonin 5-HT2A receptors. The combination has been studied for enhanced efficacy in treating depressive symptoms, particularly where monotherapy with duloxetine may be insufficient or when managing comorbid anxiety or psychotic features. Clinical studies suggest this combination can improve depressive and anxiety symptoms more effectively than duloxetine alone, with no significant increase in adverse effects[2][3]. Additionally, low-dose olanzapine may reduce the occurrence and severity of duloxetine-induced nausea and vomiting[3][6]. Both drugs act centrally to modulate neurotransmitter systems involved in mood regulation.
