Duocotexin is a fixed-dose combination antimalarial drug containing dihydroartemisinin and piperaquine, manufactured by Holley Pharmaceutical. Dihydroartemisinin, an artemisinin derivative, acts rapidly by generating reactive oxygen species and carbon-centered radicals upon activation by heme within the malaria parasite, potentially inhibiting the Sarco/endoplasmic reticulum Ca2+-ATPase (PfATP6). Piperaquine is a long-acting bisquinoline that interferes with the parasite's detoxification of heme into hemozoin. This combination is indicated for the treatment of uncomplicated malaria caused by *Plasmodium falciparum* and *Plasmodium vivax*. The University of California, San Francisco (UCSF) sponsored the DPART study to investigate the pharmacokinetics and safety of Duocotexin in HIV-infected children receiving various antiretroviral therapies, addressing potential drug-drug interactions.
