Duox2 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the Dual Oxidase 2 (DUOX2) gene. DUOX2 is an enzyme belonging to the NADPH oxidase family, primarily responsible for generating hydrogen peroxide (H2O2) at the apical surface of epithelial tissues and in macrophages. This H2O2 production is critical for oxidative host defense and inflammatory signaling. In experimental models, Duox2 siRNA has been used to demonstrate that DUOX2 is essential for house dust mite-induced cytokine production in keratinocytes and for the shedding of membrane-bound fractalkine in macrophages. It also plays a major role in basal H2O2 synthesis in airway epithelia. While currently utilized primarily as a research tool to study oxidative stress and innate immunity, it represents a potential therapeutic approach for modulating inflammatory diseases.
