DuRan-Bis is a novel bi-specific fusion protein designed to simultaneously inhibit programmed death-ligand 1 (PD-L1) and vascular endothelial growth factor (VEGF). It utilizes a single polypeptide chain scFv-scFv-Fc scaffold, where a high-affinity single-chain variable fragment (scFv) targeting PD-L1 is fused to an scFv targeting VEGF, which is in turn attached to an Fc fragment. This dual-targeting approach aims to synergistically enhance anti-cancer effects by reversing immune suppression (via PD-L1 blockade) and inhibiting tumor angiogenesis and proliferation (via VEGF blockade). Preclinical studies have demonstrated its efficacy in inhibiting the proliferation of glioblastoma cells and decreasing the viability of head and neck cancer cells when combined with immune cells.
