Overview

Durvalumab + rituximab + cyclophosphamide + doxorubicin + vincristine + prednisone is a combination regimen studied for the treatment of high-risk diffuse large B-cell lymphoma (DLBCL). It combines **durvalumab**, a monoclonal antibody immune checkpoint inhibitor targeting PD-L1; **rituximab**, a monoclonal antibody targeting CD20 on B cells; **cyclophosphamide**, an alkylating chemotherapy agent; **doxorubicin**, an anthracycline chemotherapy agent; **vincristine**, a vinca alkaloid disrupting microtubule formation; and **prednisone**, a corticosteroid with immunosuppressive and antitumor effects. This regimen aims to leverage both immune checkpoint blockade and cytotoxic chemotherapy for improved antitumor response. Clinical evidence suggests feasibility and no new safety signals, but no significant efficacy improvement over standard R-CHOP[1].
