Durvalumab + cisplatin + etoposide is a combination regimen used as a first-line treatment for adults with extensive-stage small cell lung cancer (ES-SCLC). Durvalumab is a human monoclonal antibody targeting PD-L1 (Programmed death-ligand 1), functioning as an immune checkpoint inhibitor to enhance antitumor immune responses. Cisplatin is a platinum-based chemotherapeutic that induces DNA crosslinking and apoptosis, while etoposide is a topoisomerase II inhibitor, inhibiting DNA replication and subsequently leading to cancer cell death. This combination is administered intravenously and has demonstrated a significant improvement in overall survival in ES-SCLC compared to chemotherapy alone. The combination regimen is typically administered every 21 days for 4 cycles, followed by durvalumab maintenance every 28 days until disease progression or unacceptable toxicity[1][2][4][5][7][9].
