Drug intelligence / Profile preview
Durvalumab + datopotamab deruxtecan is a combination of two anticancer agents under investigation primarily for solid tumors, including breast and lung cancer. Durvalumab is a fully human IgG1 monoclonal antibody that targets and blocks PD-L1, inhibiting its interaction with PD-1 and CD80, thereby enhancing anticancer T-cell immune responses. Datopotamab deruxtecan (Dato-DXd) is an antibody-drug conjugate (ADC) consisting of a humanized monoclonal antibody that targets TROP2 (trophoblast cell-surface antigen 2) linked to the cytotoxic payload DXd, a DNA topoisomerase I inhibitor, via a tumor-selective cleavable linker. The combination seeks to harness the immunomodulatory effect of durvalumab and the targeted cytotoxicity of Dato-DXd, with preclinical data suggesting that topoisomerase I inhibition may further stimulate antitumor immune responses. Clinical trials have shown promising activity and a manageable safety profile in indications such as triple-negative breast cancer and non-small cell lung cancer[1][3][4][5][6][7].
