Overview

Durvalumab + domvanalimab is an investigational combination therapy for cancer, primarily being studied in **unresectable, locally advanced non-small cell lung cancer (NSCLC) following platinum-based chemoradiotherapy**[1][3][5][6][8]. **Durvalumab** is a human IgG1 monoclonal antibody targeting *programmed death-ligand 1 (PD-L1)*, acting as a checkpoint inhibitor to restore T-cell anti-tumor activity. **Domvanalimab** (AB154) is an Fc-silent humanized IgG1 monoclonal antibody targeting *T-cell immunoreceptor with Ig and ITIM domains (TIGIT)*, blocking its interaction with CD112 and CD155, thereby reducing T-cell and natural killer cell inhibition and promoting anti-tumor activity[5][8]. The combination aims to synergize PD-L1 and TIGIT blockade, enhancing immune response against tumors. This therapy is currently investigational and has not received FDA approval as of September 2025[3].
