Durvalumab + gefitinib is an investigational combination therapy consisting of two distinct agents used in oncology. Durvalumab is a selective, high-affinity human IgG1κ monoclonal antibody that blocks programmed cell death ligand 1 (PD-L1) from binding to its receptors PD-1 and CD80 on T cells, thereby enhancing anti-tumor immune responses. Gefitinib is a small molecule tyrosine kinase inhibitor targeting the epidermal growth factor receptor (EGFR), inhibiting downstream signaling pathways involved in tumor cell proliferation and survival. The combination has been studied primarily in patients with advanced non-small cell lung cancer (NSCLC) harboring activating EGFR mutations who are tyrosine kinase inhibitor–naive. Clinical trials have shown that while the regimen demonstrates anti-tumor activity, it is associated with higher toxicity—particularly elevated liver enzymes—than either agent alone. No significant improvement in progression-free survival was observed compared to historical controls for monotherapy[1][3][5][6].
