Durvalumab + mocetinostat is an investigational combination therapy for cancer, specifically advanced or metastatic solid tumors and non-small cell lung cancer (NSCLC). Durvalumab is a human monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 to counteract tumor immune evasion. Mocetinostat is a selective inhibitor of class I/IV histone deacetylases (HDACs), which can modulate gene expression and potentially enhance the efficacy of immune checkpoint inhibitors like durvalumab by increasing tumor immunogenicity. The combination aims to overcome resistance to checkpoint inhibitors in NSCLC, including in patients previously unresponsive to anti-PD-(L)1 therapies[1][2][4].
