Drug intelligence / Profile preview
Durvalumab + olaparib is a combination of two pharmaceutical agents used in oncology clinical trials. Durvalumab is a human monoclonal antibody that targets programmed cell death ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80, thereby promoting antitumor immune responses by enabling cytotoxic T cells to attack tumor cells. Olaparib is a small molecule inhibitor of poly [ADP-ribose] polymerase (PARP), an enzyme involved in DNA repair; by inhibiting PARP, olaparib induces synthetic lethality in cancer cells with homologous recombination repair deficiencies, such as those harboring BRCA mutations. This combination has been investigated for synergistic effects in various cancers, including breast cancer, endometrial cancer, ovarian cancer, and small cell lung cancer[2][3][5][6]. The rationale for the combination is that PARP inhibition may increase tumor immunogenicity and sensitize tumors to immune checkpoint blockade.
