Overview

Durvalumab + olaparib + paclitaxel is a combination therapy that brings together three distinct mechanisms of action. Durvalumab is a PD-L1 inhibitor (checkpoint inhibitor), olaparib is a PARP inhibitor, and paclitaxel is a taxane chemotherapy agent. The combination was designed to boost immune response by adding an anti-PD-1 checkpoint inhibitor, as PARP inhibitors are known to increase the expression of PD-L1 protein in tumors. This triplet regimen has shown improved pathological complete response rates in HER2-negative breast cancer, particularly in triple-negative breast cancer and a subset of hormone receptor-positive/HER2-negative patients with MammaPrint ultra-high (MP2) status. The combination has also demonstrated efficacy in advanced endometrial cancer.
