Overview

This is a combination therapy consisting of durvalumab (an anti-PD-L1 monoclonal antibody), oleclumab (an anti-CD73 monoclonal antibody), and standard cytotoxic chemotherapy agents. Durvalumab blocks the programmed death-ligand 1 (PD-L1) pathway, enhancing T cell-mediated anti-tumor immunity. Oleclumab inhibits CD73, thereby reducing adenosine-mediated immunosuppression within the tumor microenvironment and further promoting anti-tumor immune responses. Chemotherapy agents widely used in these combinations include platinum-based drugs (such as oxaliplatin or cisplatin), gemcitabine, nab-paclitaxel, mFOLFOX, and others depending on the cancer indication. This triple combination is being investigated in multiple cancers, including metastatic microsatellite-stable colorectal cancer, non-small cell lung cancer, and pancreatic cancer, for its synergistic antitumor efficacy[1][2][4][5].
