Overview

Durvalumab + pomalidomide is a combination therapy studied in patients with relapsed/refractory multiple myeloma. **Durvalumab** is a monoclonal antibody that inhibits **programmed death ligand 1 (PD-L1)**, thereby blocking an immune checkpoint pathway and promoting antitumor immune responses. **Pomalidomide** is an immunomodulatory agent (IMiD) that enhances immune system activity against myeloma via multiple mechanisms, including stimulating T-cells and natural killer (NK) cells and inhibiting tumor-supportive microenvironment signals. This combination was investigated to harness additive or synergistic immunomodulation, with clinical studies showing modulation of immune cell subsets and gene expression profiles in the disease microenvironment[1][2].
