Overview
Durvalumab + trabectedin is an investigational combination therapy consisting of durvalumab, a monoclonal antibody immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1), and trabectedin, a DNA-binding small molecule chemotherapeutic agent. Durvalumab blocks the interaction between PD-L1 and its receptors (PD-1 and CD80), thereby enhancing anti-tumor immune responses. Trabectedin binds to the minor groove of DNA, disrupting cell division and modulating the tumor microenvironment by affecting tumor-associated macrophages. This combination has been studied in advanced pretreated soft tissue sarcoma (STS) and platinum-resistant/refractory ovarian carcinoma, showing manageable safety profiles and preliminary evidence of anti-tumor activity[2][3][4].
