Overview

Durvalumab plus tremelimumab in combination with platinum-based chemotherapy is an FDA-approved regimen for the first-line treatment of adult patients with metastatic non-small cell lung cancer (NSCLC) without sensitizing EGFR or ALK mutations. Durvalumab is a monoclonal antibody targeting programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 to enhance anti-tumor immune responses. Tremelimumab is a monoclonal antibody that targets cytotoxic T-lymphocyte–associated antigen 4 (CTLA-4), promoting T-cell activation and proliferation. The addition of platinum-based chemotherapy further enhances anti-cancer efficacy by directly killing rapidly dividing tumor cells. This combination was approved based on improved overall survival demonstrated in the phase III POSEIDON trial. Both durvalumab and tremelimumab are developed by AstraZeneca[1][5][7].
