**Durvalumab + tremelimumab + lenvatinib** is an experimental combination regimen being investigated for the treatment of unresectable hepatocellular carcinoma. Durvalumab is a human monoclonal antibody that blocks PD-L1, enhancing anti-tumor T-cell responses. Tremelimumab is a human monoclonal antibody targeting CTLA-4, also enhancing immune activation against tumors. Lenvatinib is an oral small molecule inhibitor targeting multiple receptor tyrosine kinases, including VEGFR, FGFR, PDGFRα, RET, and KIT, impeding tumor angiogenesis and proliferation. This combination aims to synergistically stimulate immune-mediated anti-tumor effects while inhibiting angiogenesis. The combination is being tested in several phase II and III clinical trials in patients with hepatocellular carcinoma, typically for those with previously untreated unresectable disease[2][3][5][7][9][10].
