Overview

This is a combination regimen of three drugs used in oncology clinical trials: - **Durvalumab** is a human monoclonal antibody that targets programmed cell death ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 to promote antitumor immune responses by cytotoxic T cells. - **Tremelimumab** is a human monoclonal antibody targeting cytotoxic T lymphocyte-associated antigen 4 (CTLA-4), enhancing immune activation against tumor cells. - **Paclitaxel** is a small molecule chemotherapeutic agent that stabilizes microtubules and inhibits cell division. The combination has been studied in advanced cancers such as biliary tract carcinoma and metastatic gastric cancer after failure of platinum-based chemotherapy. The rationale for combining these agents is to enhance the immune-mediated attack on tumors while leveraging the cytotoxic effects of paclitaxel. However, safety concerns have arisen regarding increased risk of severe anaphylactic reactions when all three are combined[5][8].
