Overview

Durvalumab + tremelimumab + paclitaxel is an investigational combination regimen evaluated for the treatment of metastatic gastric cancer (mGC). The regimen combines two immune checkpoint inhibitors—durvalumab, a monoclonal antibody targeting programmed cell death ligand 1 (PD-L1), and tremelimumab, a monoclonal antibody targeting cytotoxic T-lymphocyte-associated protein 4 (CTLA-4)—with paclitaxel, a taxane-based chemotherapy agent that inhibits microtubule disassembly. This triplet therapy was investigated in a Phase Ib/II clinical trial (NCT03751761) as a second-line treatment for patients who failed first-line chemotherapy, particularly those with specific biomarkers such as MSI-high or high tumor mutation burden. The study was sponsored by Asan Medical Center with drugs provided by AstraZeneca.
