Overview

Durvalumab + vistusertib is an experimental combination therapy composed of durvalumab, a monoclonal antibody targeting programmed death-ligand 1 (PD-L1) and vistusertib, an oral small molecule dual inhibitor of mammalian target of rapamycin complex 1 and 2 (mTORC1/2). Durvalumab functions as an immune checkpoint inhibitor that enhances anti-tumor immune responses by blocking PD-L1, thereby restoring T-cell activity. Vistusertib inhibits mTORC1/2, important regulators of cell growth and proliferation, aiming to overcome resistance to immune checkpoint therapies. This combination is under investigation for the treatment of advanced solid tumors, particularly non-small cell lung cancer (NSCLC), with studies designed as biomarker-directed or with molecular stratification.[1][3][8]
