Overview

Dusigitumab is a fully human monoclonal antibody developed for the treatment of cancer. It binds and neutralizes insulin-like growth factors IGF1 and IGF2, thereby inhibiting their activation of membrane-bound IGF receptors (IGF1R and IR-A). This blockade disrupts downstream signaling pathways involved in cell proliferation, survival, and oncogenic transformation, potentially leading to reduced tumor growth and increased apoptosis. Dusigitumab was originally developed by MedImmune (acquired by AstraZeneca) using Xenomouse technology licensed from Abgenix. Clinical development targeted advanced solid malignancies including unresectable or metastatic hepatocellular carcinoma and hormone-sensitive, HER2-negative metastatic breast cancer; however, all development has been discontinued due to safety and efficacy issues[1][2][3][4][6].
