Duterporfin is a second-generation porphyrin-based photosensitizer developed by Shanghai Fudan-Zhangjiang Bio-Pharmaceutical for use in photodynamic therapy (PDT). It is primarily being investigated for the treatment of advanced hilar cholangiocarcinoma. The drug is administered intravenously and selectively accumulates in tumor tissues. Upon activation by a specific wavelength of laser light (typically delivered via endoscopic retrograde cholangiopancreatography), duterporfin reacts with molecular oxygen to produce cytotoxic reactive oxygen species, such as singlet oxygen. These species induce oxidative damage, leading to tumor cell apoptosis, necrosis, and microvascular occlusion, thereby reducing tumor burden and improving biliary patency.
