Duvakitug is a human IgG1-λ2 monoclonal antibody that targets tumor necrosis factor (TNF)-like ligand 1A (TL1A), also known as TNF superfamily member 15 (TNFSF15). TL1A signaling amplifies inflammation and drives fibrosis in inflammatory bowel disease (IBD) by binding to its receptor DR3. By inhibiting TL1A signaling via DR3, duvakitug aims to reduce the overactive immune response and fibrotic processes associated with IBD. Duvakitug has demonstrated efficacy and favorable safety as an induction treatment in adults with moderately to severely active ulcerative colitis and Crohn’s disease in phase 2b clinical trials. It is being developed through a collaboration between Teva Pharmaceutical Industries and Sanofi, with plans for phase 3 development pending regulatory discussions[1][2][4][6][7].
