duvelisib

Duvelisib is an oral small molecule inhibitor of phosphoinositide 3-kinase (PI3K), specifically targeting the delta and gamma isoforms. It is indicated for the treatment of adults with relapsed or refractory chronic lymphocytic leukemia (CLL), small lymphocytic lymphoma (SLL), and follicular lymphoma after at least two prior therapies have failed. By inhibiting PI3K-delta, duvelisib directly impairs proliferation and survival of malignant B-cells; inhibition of PI3K-gamma disrupts cytokine signaling and reduces support from the tumor microenvironment. This dual mechanism suppresses both cancer cell growth and their supportive immune environment[3][5][6][8]. Duvelisib was originally developed by Infinity Pharmaceuticals and later by Verastem.
