A combination investigational therapy comprising **duvortuxizumab**, a humanized bispecific single-chain variable fragment (scFv) antibody that targets CD19 and CD3, and **ibrutinib**, a first-in-class, orally available, covalent inhibitor of Bruton’s tyrosine kinase (BTK). Duvortuxizumab redirects T cells to target and kill malignant B cells by engaging both CD19 on B cells and CD3 on T cells, acting as a T-cell redirecting agent. Ibrutinib inhibits BTK, a key enzyme in the B-cell receptor signaling pathway, leading to reduced proliferation and survival of B-cell malignancies. The combination is under clinical investigation for safety, tolerability, and efficacy, particularly in B-cell malignancies such as relapsed/refractory non-Hodgkin lymphoma.
