DVD-3E is an experimental logic-gated T-cell engager (TCE) linked with a mutant interleukin-2 (IL-2) designed to treat solid tumors while minimizing systemic toxicity. The construct utilizes a protease-activatable dual-variable-domain (DVD) format, acting as a 'Pro-TCE' that remains inert in systemic circulation to prevent off-tumor toxicity. Upon exposure to tumor-specific proteases within the tumor microenvironment (TME), the construct is cleaved and activated. The linked mutant IL-2, designated '3E', has markedly reduced receptor affinity to ensure it is inactive in the periphery and only provides 'cis' signaling to T cells engaged by the TCE component. This design principle aims to enhance the persistence and effector function of intratumoral T cells, expand progenitor T cell populations, and establish durable immune memory without the severe side effects associated with systemic IL-2 or conventional TCE therapies.
