Overview

DW-4421 is a small molecule potassium-competitive acid blocker (P-CAB) being developed by Daewon Pharm for the treatment of gastroesophageal reflux disease (GERD). Unlike traditional proton pump inhibitors (PPIs) that require activation by gastric acid and bind covalently to the H+/K+-ATPase, P-CABs like DW-4421 competitively and reversibly bind to the potassium-binding site of the gastric proton pump. This mechanism allows for a more rapid onset of action, acid-independent activation, and more sustained suppression of gastric acid secretion, potentially addressing the limitations of PPIs such as nocturnal acid breakthrough and slow onset of relief.
