It acts as a dual covalent inhibitor targeting both Bruton's tyrosine kinase (BTK) and interleukin-2-inducible T-cell kinase (ITK), which are key regulators in the activation of B cells and T cells. By inhibiting both BTK and ITK simultaneously, DWP213388 offers broader immunomodulatory effects compared to agents that target only one cell type. Preclinical studies have demonstrated its potent efficacy in models of autoimmune disease such as rheumatoid arthritis. The drug has shown high selectivity for its targets with minimal off-target activity against kinases like EGFR. Developed by Daewoong Pharmaceutical, it has entered phase 1 clinical trials for chronic urticaria and was previously investigated for other autoimmune indications including rheumatoid arthritis, systemic lupus erythematosus, graft-versus-host disease, and psoriasis[1][3][5][6][8].
